Abstract

As one of the world's major food crops, the research and improvement of rice traits and breeding have always been a field of concern in agricultural science. Since the 20th century, the increasingly mature gene editing technology, from ZFNs technology to CRISPR, has been increasingly applied in the improvement of rice varieties. This research summarizes some application methods of gene editing technology, mainly CRISPR technology, in rice variety improvement by summarizing relevant articles, such as cutting, knocking out, or precise replacement of specified genes; And other related biotechnology that can be combined with CRISPR technology in the practical application of rice gene editing were introduced, including but not limited to subcellular localization analysis, gene overexpression technology, and a collection of gene diagnosis and editing. Meanwhile, for some of the remaining issues with CRISPR technology, such as gene deletions, accidental insertions, and chromosomal aberrations caused by target DNA sequence disruption, a comprehensive evaluation of the effectiveness of gene editing is conducted through composite analysis of clone gene typing, PCR related detection, single molecule real-time sequencing, and long amplicon sequencing. This research aims to summarize the current applications and challenges of gene editing technology in rice trait research and variety improvement, and to provide prospects for its integration with other biotechnology.
